Novel Members of the AKH/RPCH Peptide Family: Isolation of AKH from the Corpora Cardiaca of the Two Beetle Species, Cheilomenes Lunata and Coccinella Septempunctata

چکیده

Adipokinetic hormones (AKH) are synthesized in endocrine cells of the glandular corpora cardiaca, in the anterior part of the retrocerebral complex. AKH plays an important role in the metabolism of carbohydrates that fuel energydepleting activities such as flight and walking. To date, the primary sequences of eight isoforms from various beetle species have been elucidated. In this study, the glandular part of the CC from two Ladybird beetles, Cheilomenes lunata and Coccinella septempunctata were directly analysed by MALDI-TOF mass spectrometry. Subsequently, the amino acid sequences of both putative novel AKHs were confirmed by tandem mass spectrometry.

Flight substrates and their regulation by a member of the AKH/RPCH family of neuropeptides in Cerambycidae.

The pattern of metabolic changes during tethered flight with lift-generation was investigated in two South African species of long-horned beetles (family: Cerambycidae), namely Phryneta spinator and Ceroplesis thunbergi. Energy substrates were measured in haemolymph and flight muscles at rest, after a flight period of 1 min at an ambient temperature of 25-29 degrees C, and 1 h thereafter. Fligh...

A third active AKH is present in the pyrgomorphid grasshoppers Phymateus morbillosus and Dictyophorus spumans.

The corpora cardiaca of the African pyrgomorphid grasshoppers Phymateus morbillosus and Dictyophorus spumans contain three adipokinetic hormones (AKHs): besides two already known AKHs, Phm-AKH-I and Scg-AKH-II (Gäde et al., 1996 [Gäde, G., Kellner, R., Rinehart, K.L., 1996. Pyrgomorphid grasshoppers of the genus Phymateus contain species-specific decapeptides of the AKH/RPCH family regulating l...

A new member of the AKH/RPCH family that stimulates locomotory activity in the firebug, Pyrrhocoris apterus (Heteroptera).

A new member of the AKH/RPCH family was isolated and identified from the corpora cardiaca of the firebug Pyrrhocoris apterus. The peptide was isolated in a single step by reversed phase HPLC and the structure deduced from the multiple MS (MS(N)) electrospray mass spectra and amino acid analysis as that of an octapeptide with the sequence pGlu-Leu-Asn-Phe-Thr-Pro-Asn-Trp-NH(2): this sequence was...

Isolation and structure of a novel charged member of the red-pigment-concentrating hormone-adipokinetic hormone family of peptides isolated from the corpora cardiaca of the blowfly Phormia terraenovae (Diptera).

A hypertrehalosaemic neuropeptide from the corpora cardiaca of the blowfly Phormia terraenovae has been isolated by reversed-phase h.p.l.c., and its primary structure was determined by pulsed-liquid phase sequencing employing Edman chemistry after enzymically deblocking the N-terminal pyroglutamate residue. The C-terminus was also blocked, as indicated by the lack of digestion when the peptide ...

The adipokinetic hormone family in Chrysomeloidea: structural and functional considerations *

The presented work is a hybrid of an overview and an original research paper on peptides belonging to the adipokinetic hormone (AKH) family that are present in the corpora cardiaca of Chrysomeloidea. First, we introduce the AKH/red pigment-concentrating hormone (RPCH) peptide family. Second, we collate the available primary sequence data on AKH peptides in Cerambycidae and Chrysomelidae, and we...
